Human Heparanase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com designed for the in vitro quantitative determination of human Heparanase 1 in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 × 8 well strips and produces a colorimetric signal read at 450 nm, with a TMB substrate generating a blue product that turns yellow after addition of acidic stop solution; the absorbance is proportional to the concentration of Heparanase 1 captured in each well.

The reported sensitivity is 11.3 pg/ml with a product range of 31.2–2000 pg/ml, and the assay is completed in 4 hours 30 minutes. Recovery in spiked samples averaged 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5), with linearity data reported for serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions. Heparanase 1 in standards and samples is captured by the immobilised antibody and detected with a biotinylated anti-Heparanase 1 antibody and HRP-Avidin conjugate, with target concentration calculated from a standard curve.
