Human Hedgehog Homolog, Sonic ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) enables the in vitro quantitative determination of human Sonic Hedgehog in tissue homogenates, cell lysates or other biological fluids. An antibody specific for Sonic Hedgehog is pre-coated onto the wells of a microplate configured as 12 x 8 strips; standards and samples are added, the captured analyte is detected with a biotinylated anti-Sonic Hedgehog antibody followed by an HRP-Avidin conjugate, and the reaction is visualised with TMB substrate, producing a blue product that converts to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the concentration is derived from the standard curve.

Working range is 78.1-5000 pg/ml and sensitivity is 31 pg/ml, with a total run time of 4h 30m. Kit components are stored at +4°C or -20°C as indicated and include a pre-coated 96-well microplate, lyophilised standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Spike-and-recovery data (n=5) are provided for serum (range 80%-102%, average 91%), EDTA plasma (range 81%-100%, average 90%) and heparin plasma (range 80%-89%, average 84%).
