Antibodies.com provides a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of human Heat Shock Protein Beta 7 (HSPB7) in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with bound HSPB7 detected using a biotinylated anti-HSPB7 antibody and an HRP-avidin conjugate. A TMB substrate produces a colour change that is read at 450 nm, and the total assay time is approximately 4 hours 30 minutes.
According to the manufacturer, sensitivity is <0.124 ng/ml with a quantitative range of 0.312–20 ng/ml. Reported recovery in spiked samples averages 91% in serum, 89% in EDTA plasma and 90% in heparin plasma (n=5 per matrix). Linearity testing across 1:2, 1:4, 1:8 and 1:16 dilutions gave ranges of 87–109%, 74–104%, 90–94% and 86–108% for serum; 86–101%, 90–101%, 78–110% and 84–96% for EDTA plasma; and 90–101%, 77–107%, 93–96% and 89–109% for heparin plasma. Kit components include a pre-coated microplate, two vials of lyophilized standard, detection solutions A and B, a 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or –20°C as instructed.
