Human Heat Shock Protein Beta 3 ELISA Kit

From Antibodies.com, this Human HSPB3 (Heat Shock Protein Beta 3) ELISA Kit is a sandwich enzyme-linked immunosorbent assay designed for the in vitro quantitative determination of human HSPB3 in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microtiter plate configured as 12 x 8 well strips, with detection via a biotinylated anti-HSPB3 antibody and an HRP-avidin conjugate, visualised with TMB substrate and read at 450 nm on a colorimetric microplate reader.

Reported sensitivity is <0.29 ng/ml and the assay range is 0.781–50 ng/ml, with a total assay time of 4h 30m. The kit is validated for serum, EDTA plasma, and heparin plasma. Average recovery across these sample types (n=5) ranged from 80–99%, with reported averages of 91% for serum, 89% for EDTA plasma, and 90% for heparin plasma. Linearity at dilutions of 1:2, 1:4, 1:8, and 1:16 was reported as 87–109%, 74–104%, 90–94%, and 86–108% for serum (n=5); 86–101%, 90–101%, 78–110%, and 84–96% for EDTA plasma (n=5); and 90–101%, 77–107%, 93–96%, and 89–109% for heparin plasma (n=5).

Kit components include a pre-coated microplate, lyophilized standard (2 vials), detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and plate sealers, stored at +4°C or -20°C as instructed. Tissue samples are homogenised in PBS (1g tissue / 9ml PBS) and centrifuged at 5000 x g for 5 minutes, with total protein per well not exceeding 0.3mg.
