Description

Designed by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) enables the in vitro quantitative determination of human Heat Shock Protein Beta 2 (HSPB2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for HSPB2. Standards and samples are added to the wells, where HSPB2 binds to the immobilized capture antibody. After washing, a biotinylated anti-HSPB2 detection antibody is added, followed by HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue-coloured product that turns yellow upon addition of acidic stop solution, with colour intensity directly proportional to HSPB2 concentration. Absorbance is read at 450 nm on a colorimetric microplate reader.

The kit has a measuring range of 0.625–40 ng/ml and a sensitivity of <0.244 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Spike-and-recovery testing across five samples per matrix yielded average recoveries of 91% for serum (range 83–99%), 89% for EDTA plasma (range 80–99%), and 90% for heparin plasma (range 82–99%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma samples. Components should be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
