Description

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human HSP90 beta in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit uses a pre-coated microplate format and employs the sandwich enzyme immunoassay technique, in which an antibody specific for HSP90 beta is immobilised on the plate to capture target from standards and test samples, followed by incubation with a Biotinylated Anti-HSP90 beta Antibody and an HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450 nm in a microplate reader; the yellow intensity is proportional to HSP90 beta concentration.

The standard curve spans 1.56–100 ng/ml, with a reported sensitivity of 0.67 ng/ml and an assay time of 4 hours 30 minutes. Reagent components are stored at +4°C or −20°C as instructed.

Spike-and-recovery performance was 80–102% (average 91%) in serum (n=5), 81–101% (average 91%) in EDTA plasma (n=5) and 80–89% (average 84%) in heparin plasma (n=5). Linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. The kit is for research use only and is not intended for diagnostic or therapeutic use.
