Human Heat Shock 70kDa Protein 1 Like Protein ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com and intended for the in vitro quantitative determination of human HSPA1L in tissue homogenates or other biological fluids. An antibody specific for HSPA1L is pre-coated onto a 96-well microplate; standards and test samples are added and HSPA1L is bound by the immobilised antibody. After incubation and washing, a Biotinylated Anti-HSPA1L Antibody and HRP-Avidin conjugate are added, and TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow on addition of acidic stop solution. Yellow intensity is proportional to HSPA1L captured in each well, and concentration is calculated from the O.D. absorbance at 450 nm read on a microplate reader against the standard curve.

The kit reports a sensitivity of 0.063 ng/ml and an assay range of 0.156–10 ng/ml. Total assay time is 4 h 30 min, run on a pre-coated microplate of 12 x 8 well strips with colourimetric detection. Sample recovery has been demonstrated in serum (n = 5, range 83%–100%, average 91%), EDTA plasma (n = 5, range 90%–98%, average 94%) and heparin plasma (n = 5, range 89%–102%, average 95%). Across 1:2, 1:4, 1:8 and 1:16 dilutions, recovery ranges were 83–110%, 79–100%, 90–98% and 82–108% for serum (n = 5); 90–98%, 90–105%, 73–109% and 87–91% for EDTA plasma (n = 5); and 88–103%, 77–105%, 95–98% and 87–110% for heparin plasma (n = 5).

Components supplied are a pre-coated 96 well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers.
