Human Guanylate Cyclase Activator 2B ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Uroguanylin (Guanylate Cyclase Activator 2B) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated microplate (12 x 8 well strips) with a colourimetric detection format. An antibody specific for Uroguanylin is immobilised on the plate; captured analyte is detected using a Biotinylated Anti-Uroguanylin Antibody followed by an HRP-Avidin conjugate and TMB substrate, producing a yellow product read at 450 nm in a microplate reader.

The standard assay range is 15.6-1000 pg/ml with a sensitivity of <6.2 pg/ml and a total assay time of 4 hours 30 minutes. Kit components include the pre-coated microplate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers; these are stored at +4°C or -20°C as instructed. Reported recovery averaged 91% in serum (range 83%-100%), 94% in EDTA plasma (range 90%-98%) and 94% in heparin plasma (range 90%-99%).

Materials required but not included are a microplate reader capable of measuring absorbance at 450 nm ± 10 nm, a multichannel pipette reservoir or automated microplate washer, software capable of generating logarithmic functions, test tubes and Eppendorf tubes, multi- and single-channel pipettes with sterile tips, deionized or distilled water, absorbent paper towels, and a 37°C incubator.
