Human Growth Factor Receptor Bound Protein 2 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is designed for the in vitro quantitative determination of human Growth Factor Receptor Bound Protein 2 (GRB2) in tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique: an antibody specific for GRB2 is pre-coated onto a microplate, standards and test samples are added, and GRB2 is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-GRB2 Antibody binds the captured GRB2, then an HRP-Avidin conjugate is added. A TMB substrate solution is used to visualize the reaction, producing a blue product that changes to yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of GRB2 captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The assay is performed on a pre-coated microplate (12 x 8 well strips) with a total assay time of 4h 30m and is intended for use with a colorimetric microplate reader. Sample types include tissue homogenates or other biological fluids, with reactivity confirmed for human samples. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Reported performance characteristics include a sensitivity of 0.055 ng/ml and an assay range of 0.156–10 ng/ml. Average recovery values from n=5 samples are 91% for serum (range 83%–99%), 89% for EDTA plasma (range 80%–99%), and 90% for heparin plasma (range 82%–99%). Linearity has been evaluated at 1:2 to 1:16 dilutions for serum, EDTA plasma, and heparin plasma samples.
