Human Growth Differentiation Factor 9 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com designed for the in vitro quantitative determination of human Growth Differentiation Factor 9 (GDF9). The assay is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. A 96-well microplate is pre-coated with an antibody specific for GDF9; standards and samples are added and any GDF9 present is captured by the immobilised antibody. After washing, a biotinylated anti-GDF9 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that turns yellow on addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the GDF9 concentration is calculated from the standard curve.

The measurement range is 0.156–10 ng/ml with a sensitivity of 0.067 ng/ml. The assay takes approximately 4h 30m to complete. Spike-and-recovery performance has been validated in serum (n=5, range 80–102%, average 91%), EDTA plasma (n=5, range 81–100%, average 90%) and heparin plasma (n=5, range 80–89%, average 84%). Dilution linearity has also been characterised at 1:2, 1:4, 1:8 and 1:16 dilutions for each of these sample matrices.

Detection is colorimetric and the assay is intended for use with human samples. Kit components are stored at +4°C or -20°C as instructed in the protocol manual and the kit should not be used past its expiration date.
