Human Growth Differentiation Factor 5 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com for the in vitro quantitative determination of human Growth Differentiation Factor 5 (GDF-5). The kit is intended for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay uses a microplate pre-coated with an antibody specific for GDF-5. Standards and samples are added to the wells, captured GDF-5 is bound by a Biotinylated Anti-GDF-5 Antibody, and an HRP-Avidin conjugate is added. Detection is colourmetric, using TMB substrate with absorbance read at 450 nm on a microplate reader. The standard curve covers a product range of 0.156–10 ng/ml and the reported sensitivity is 0.064 ng/ml. The total assay time is approximately 4 h 30 min, and components are stored at +4 °C or −20 °C as instructed; the kit should not be used past its expiration date.

Spike-and-recovery testing (n = 5) returned averages of 91 % for serum (range 80–102 %), 90 % for EDTA plasma (81–100 %) and 84 % for heparin plasma (80–89 %). Dilution linearity was demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions, with serum ranging from 74 % to 107 %, EDTA plasma from 79 % to 108 % and heparin plasma from 82 % to 105 % across the series.
