Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Growth Differentiation Factor 1 (GDF1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for GDF1, with captured protein detected using a Biotinylated Anti-GDF1 Antibody and an HRP-Avidin conjugate. The HRP enzymatic reaction is visualised with TMB substrate solution, producing a blue-coloured product that turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of GDF1 captured in each well, and the concentration is calculated from the O.D. absorbance read at 450 nm on a colorimetric microplate reader.

The kit provides a quantitative measurement range of 31.2-2000 pg/ml with a sensitivity of <10.2 pg/ml, and the complete assay can be performed in 4h 30m. In spike-and-recovery testing (n=5), average recoveries were 91% for serum (range 80%–102%), 90% for EDTA plasma (range 81%–100%) and 84% for heparin plasma (range 80%–89%). Linearity upon dilution was demonstrated at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma samples.

Kit components include a pre-coated 96 well microplate arranged as 12 x 8 well strips, lyophilised standard (2 vials), Detection Solution A and Detection Solution B (120 μl each), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Storage conditions vary by component: the microplate, lyophilised standard and detection solutions should be kept at -20°C, while the wash buffer, TMB substrate and stop solution are stored at +4°C; the kit should not be used past its expiration date.
