Description

Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human Granzyme K in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit is read on a colorimetric microplate reader and is built around a pre-coated 96-well microplate arranged as 12 x 8 well strips.

The assay uses the sandwich enzyme immunoassay technique. Standards and samples are added to the antibody-coated wells and Granzyme K present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-Granzyme K Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of Granzyme K captured and is quantified by reading the O.D. absorbance at 450 nm against a standard curve. The reported product range is 31.2-2000 pg/ml with a sensitivity of <13.1 pg/ml, and the assay is completed in 4h 30m.

Spike-and-recovery performance is reported for serum (range 80%-102%, average 91%), EDTA plasma (range 81%-100%, average 90%) and heparin plasma (range 80%-89%, average 84%). Linearity has been evaluated across dilutions of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. Components supplied include the pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at either +4°C or -20°C as indicated, and the kit should not be used past its expiration date.
