As a sandwich ELISA, an antibody specific to the target is pre-coated onto a microplate. Standards and test samples are added to the wells, where the target analyte in the sample is captured by the immobilised antibody. Following incubation and washing, a biotinylated detection antibody binds the captured target, and an HRP-avidin conjugate is then added. After further incubation and washing, TMB substrate solution is used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and target concentration is calculated by reference to a standard curve.
