From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is intended for the in vitro quantitative determination of human GNMT in tissue homogenates, cell lysates, or other biological fluids. The kit employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate, using a biotinylated anti-GNMT detection antibody followed by an HRP-avidin conjugate. TMB substrate is added to visualise the enzymatic reaction, producing a blue product that turns yellow after the acidic stop solution; the density of yellow is read as optical density at 450 nm in a microplate reader and is proportional to the amount of GNMT captured.

The assay has a reported sensitivity of 0.101 ng/ml and a working range of 0.312-20 ng/ml, with a total assay time of 4h 30m. Reported average recoveries in validation testing were 91% in serum (range 80-102%, n=5), 90% in EDTA plasma (range 81-100%, n=5), and 84% in heparin plasma (range 80-89%, n=5). Dilutional linearity across 1:2, 1:4, 1:8 and 1:16 sample dilutions was assessed in serum, EDTA plasma and heparin plasma. Standards are prepared by 1:2 serial dilution from 20 ng/ml down to 0.313 ng/ml in Sample Dilution Buffer, with the diluent serving as the zero standard.

Supplied components include a Pre-Coated 96 Well Microplate (12 x 8 well strips), Lyophilized Standard (2 vials), Detection Solution A (120μl), Detection Solution B (120μl), Wash Buffer 30X (20ml), Sample Dilution Buffer (45ml), TMB Substrate (9ml), Stop Solution (6ml), and 5 adhesive plate sealers. Reagents are stored at +4°C or -20°C as indicated on the component table. The product is for research use only and is not intended for diagnostic or therapeutic use.
