Human Glyceraldehyde-3-Phosphate Dehydrogenase ELISA Kit

Designed for the in vitro quantitative determination of human Glyceraldehyde-3-Phosphate Dehydrogenase (GAPDH), this sandwich ELISA from Antibodies.com is intended for use with tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs a 96-well microtiter plate pre-coated with an antibody specific for GAPDH. Standards and test samples are added to the wells, and GAPDH present in each sample is captured by the immobilized antibody. Following incubation and washing, a biotinylated anti-GAPDH antibody is added, followed by an HRP-avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the GAPDH concentration is calculated by reference to a standard curve.

The product range is 1.56–100 ng/ml, with a sensitivity of <0.57 ng/ml. Total assay time is approximately 4 hours 30 minutes. Kit components are stored at +4°C or –20°C as instructed in the manual and should not be used past the stated expiration date.

Validated recovery data (n=5 per matrix) show averages of 91% in serum (range 80%–102%), 90% in EDTA plasma (range 81%–100%), and 84% in heparin plasma (range 80%–89%).
