This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Glutathione S Transferase mu (GST mu) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate with a biotinylated detection antibody, HRP-avidin conjugate and TMB substrate. Absorbance is read at 450 nm on a colorimetric microplate reader, and total assay time is 4 hours 30 minutes.

According to the manufacturer, the standard curve covers 1.56–100 ng/ml with a sensitivity of <0.52 ng/ml. Reported average spike-and-recovery values across five replicates were 94% in serum (range 88–100%), 95% in EDTA plasma (range 90–101%) and 96% in heparin plasma (range 92–101%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 across the same sample types, with reported recovery windows generally falling between approximately 77% and 108%.

The assay principle uses an immobilised capture antibody to bind GST mu from standards or test samples. After washing, a biotinylated anti-GST mu antibody is added, followed by HRP-avidin. TMB substrate is then applied and the resulting blue product is converted to yellow by an acidic stop solution. The intensity of the yellow colour, proportional to the amount of GST mu captured in each well, is quantified against the standard curve.
