An antibody specific for the target is pre-coated onto the microplate wells; standards and samples are added, followed by a biotinylated detection antibody and an HRP-avidin conjugate. TMB substrate is used to visualise the reaction, producing a yellow product after the addition of stop solution, with absorbance read at 450 nm in a microplate reader. The intensity of colour is proportional to the amount of analyte captured in each well.

The assay demonstrates a sensitivity of <0.083 ng/ml and a quantitative range of 0.312–20 ng/ml, with a total assay time of 4 hours 30 minutes. Sample recovery has been validated in serum (average 91%, range 80%–102%, n=5), EDTA plasma (average 90%, range 81%–100%, n=5), and heparin plasma (average 84%, range 80%–89%, n=5). Linearity has been demonstrated at dilutions of 1:2, 1:4, 1:8, and 1:16 across serum, EDTA plasma, and heparin plasma samples.
