Human Glutamate Decarboxylase 2, Acid ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human GAD65 (Glutamate Decarboxylase 2, Acid) in tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, where GAD65 is captured by an immobilised antibody. After washing, a Biotinylated Anti-GAD65 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the GAD65 concentration is calculated by referring to the standard curve. Total assay time is 4h 30m.

The quantitative assay has a product range of 0.781–50 ng/ml with a sensitivity of <0.23 ng/ml. Reported average recovery is 91% for serum (n=5, range 80%–102%), 90% for EDTA plasma (n=5, range 81%–100%) and 84% for heparin plasma (n=5, range 80%–89%). Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
