Human Glucuronidase Beta ELISA Kit

Designed for the in vitro quantitative determination of human beta Glucuronidase/GUSB, this sandwich ELISA kit from Antibodies.com is reactive with human samples and is suitable for serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a pre-coated microplate in a 12 x 8 well strip configuration.

An antibody specific for beta Glucuronidase/GUSB is pre-coated onto the plate. Standards and test samples are added to the wells, where the analyte is captured by the immobilized antibody. Following incubation and washing, a biotinylated anti-beta Glucuronidase/GUSB antibody is added, followed by HRP-Avidin conjugate. After further incubation and washing, TMB substrate is added; the HRP-catalysed reaction produces a blue product that turns yellow on addition of acidic stop solution. The optical density at 450 nm, measured on a colorimetric microplate reader, is proportional to the amount of beta Glucuronidase/GUSB captured in each well and is quantified against a standard curve.

The kit has a sensitivity of <0.074 ng/ml and an assay range of 0.156-10 ng/ml. Total assay time is 4h 30m. Kit components are stored at +4°C or -20°C as instructed. Reported recovery (n=5) averages 96% (range 90-102%) in serum, 95% (90-101%) in EDTA plasma and 94% (91-98%) in heparin plasma. Dilution performance across 1:2, 1:4, 1:8 and 1:16 is also reported for serum (82-96%, 83-98%, 81-99%, 93-101%), EDTA plasma (88-101%, 86-95%, 90-102%, 80-93%) and heparin plasma (80-91%, 82-90%, 95-104%, 79-95%).
