From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human GBA in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate with colorimetric detection read at 450 nm in a microplate reader.

The procedure employs a biotinylated anti-GBA detection antibody followed by HRP-avidin conjugate and TMB substrate, producing a yellow-coloured product whose absorbance is proportional to the amount of GBA captured in each well. The stated assay range is 1.56–100 ng/ml, sensitivity is <0.59 ng/ml, and the total assay time is 4 hours 30 minutes. Reported average recovery values are 91% for serum (n=5, range 83–99%), 89% for EDTA plasma (n=5, range 80–99%) and 90% for heparin plasma (n=5, range 82–99%). Linearity data for dilutions of 1:2, 1:4, 1:8 and 1:16 across the same sample types are also provided.

Kit components include a pre-coated microplate (12 x 8 well strips), lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers. Components are stored at +4°C or −20°C as indicated, and the kit should not be used past its expiration date.
