Human Glucosidase Alpha, Acid ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables in vitro quantitative determination of human GAA (Glucosidase Alpha, Acid) in serum, plasma, tissue homogenates, cell lysates, and other biological fluids.

The assay uses a microtiter plate pre-coated with an antibody specific for GAA; standards and samples are added and any GAA present is captured by the immobilised antibody. After washing, a Biotinylated Anti-GAA Antibody binds the captured analyte, an HRP-Avidin conjugate is added, and TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the GAA concentration is calculated by reference to the standard curve. Reported performance specifications include a sensitivity of <0.064 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4h 30m.

Recovery has been characterised in serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, range 81%-100%, average 90%), and heparin plasma (n=5, range 80%-89%, average 84%). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across all three sample types. Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, stored at +4°C or -20°C as instructed.
