Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Glucocorticoid Induced Tumor Necrosis Factor Receptor (GITR) in serum, plasma, tissue homogenates, cell lysates or other biological fluids.

The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for GITR pre-coated onto a 96-well microtiter plate. Standards and test samples are added and the GITR present is bound by the immobilised antibody. Following washing, wells are incubated with Biotinylated Anti-GITR Antibody, then HRP-Avidin conjugate. A TMB substrate solution is used to visualise the enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow after the addition of acidic stop solution; the density of yellow is proportional to the amount of GITR captured in each well. Concentration is calculated by reading the O.D. absorbance at 450nm in a microplate reader and referring to the standard curve.

The assay has a sensitivity of 5.9 pg/ml and a detection range of 15.6-1000 pg/ml. Total assay time is 4 hours 30 minutes, with colourmetric detection on a pre-coated microplate platform. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
