Human Ghrelin ELISA Kit

This competitive Enzyme-Linked Immunosorbent Assay (cELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Ghrelin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids.

The kit employs a pre-coated 96-well microplate (12 x 8 well strips) in which Ghrelin in samples or standards competes with immobilised antigen for binding sites on a biotinylated anti-Ghrelin antibody. An HRP-Avidin conjugate and TMB substrate are then used to generate a colour change that is read at 450 nm on a colorimetric microplate reader, with the intensity of yellow colour inversely proportional to the Ghrelin concentration. The reported assay range is 0.156-10 ng/ml, sensitivity is 0.047 ng/ml, and the total assay time is 2h 30m. Kit components are stored at +4°C or -20°C as instructed.

Validated average recovery is 91% (range 80%-102%) in serum, 90% (range 81%-100%) in EDTA plasma, and 84% (range 80%-89%) in heparin plasma. Dilution linearity has been demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions, with serum recoveries of 87-91%, 87-107%, 74-101%, and 92-97%; EDTA plasma recoveries of 90-105%, 84-101%, 90-101%, and 79-108%; and heparin plasma recoveries of 84-95%, 92-105%, 82-105%, and 89-91%.
