Human GATA Binding Protein 3 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human GATA Binding Protein 3 (GATA3) in tissue homogenates, cell lysates and other biological fluids. The assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with the colorimetric end-point read as optical density at 450 nm on a standard microplate reader.

The working range is 0.312–20 ng/ml with a sensitivity of <0.135 ng/ml, and the total assay time is 4 hours 30 minutes. Standards are supplied lyophilized, and captured GATA3 is detected using a biotinylated anti-GATA3 antibody followed by an HRP-avidin conjugate and TMB substrate; the reaction is stopped with an acidic stop solution to yield a yellow product whose intensity is proportional to antigen concentration. Spike-and-recovery in serum (n=5) was 80–102% (average 91%), in EDTA plasma 81–100% (average 90%) and in heparin plasma 80–89% (average 84%). Dilutional linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 in serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%).

Components supplied include the pre-coated microplate, two vials of lyophilized standard, detection solution A (120 µl), detection solution B (120 µl), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and plate sealers, stored at +4°C or –20°C as instructed.
