From Antibodies.com, this competitive ELISA kit is designed for the in vitro quantitative determination of human Intrinsic Factor. It is reactive with human samples and is suitable for serum, plasma, tissue homogenates or other biological fluids.

The assay uses a pre-coated microplate (12 x 8 well strips) and follows a competitive immunoassay principle. Intrinsic Factor in samples or standards competes with immobilised antigen for binding sites on a biotinylated anti-Intrinsic Factor antibody. After washing, an HRP-avidin conjugate is added, and a TMB substrate produces a colour change that is read at 450 nm on a colorimetric microplate reader. The intensity of colour is inversely proportional to Intrinsic Factor concentration. Total assay time is 2 hours 30 minutes.

The quoted assay range is 3.12–200 ng/ml, with a sensitivity of <0.95 ng/ml. Reported average recovery values across n=5 samples are 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilution linearity is reported from 1:2 through 1:16 across all three sample types. Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
