Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Gastric Inhibitory Polypeptide (GIP) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit targets GIP, also referred to as glucose-dependent insulinotropic polypeptide or incretin hormone, and is reactive with human samples.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips and follows the sandwich enzyme immunoassay technique. Standards and samples are added to wells where GIP is captured by the immobilized antibody, followed by washing and incubation with a Biotinylated Anti-GIP Antibody. An HRP-Avidin conjugate is then added, and TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and GIP concentration is calculated by reference to the standard curve.

The stated product range is 78.1–5000 pg/ml with a sensitivity of 24.5 pg/ml, and total assay time is 4h 30m. Reported average recovery values are 91% for serum (n=5, range 80–102%), 90% for EDTA plasma (n=5, range 81–100%) and 84% for heparin plasma (n=5, range 80–89%). Dilution linearity is provided across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. Kit components should be stored at +4°C or −20°C as instructed and the kit must not be used past its expiration date.
