Human Galectin 9 ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Galectin 9 (Gal-9) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs an antibody specific for Galectin 9/Gal-9 pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-Galectin 9/Gal-9 Antibody, an HRP-Avidin conjugate, and TMB substrate. The enzymatic reaction produces a blue product that turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader.

Quantitative performance characteristics include a standard range of 78.1–5000 pg/ml, a sensitivity of <34 pg/ml, and a total assay time of 4 hours 30 minutes. The kit is supplied on a pre-coated microplate configured as 12 x 8 well strips. Spike-and-recovery in neat samples (n=5) gave ranges of 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma, and 80–89% (average 84%) for heparin plasma. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions spanned 74–108% across the three sample types. Kit components include lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and five adhesive plate sealers. Components should be stored at +4°C or -20°C as indicated on the kit insert.
