A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com for the in vitro quantitative determination of human G Protein Coupled Receptor 35 (GPR35) in tissue homogenates, cell lysates, or other biological fluids. The test employs a pre-coated 96-well microtiter plate (12 x 8 well strips) bearing an immobilised antibody specific for GPR35. Standards and samples are added to the wells, where GPR35 is captured by the coated antibody. Following incubation and washing, a biotinylated anti-GPR35 detection antibody is applied, followed by an HRP-avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that converts to yellow upon addition of acidic stop solution, with absorbance read at 450 nm using a colourimetric microplate reader.

Reported sensitivity is <0.068 ng/ml with an assay range of 0.156-10 ng/ml and a total assay time of 4h 30m. Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, stored at +4°C or -20°C as instructed. In spiked recovery testing (n=5), serum averaged 91% (range 83-100%), EDTA plasma averaged 94% (range 91-98%), and heparin plasma averaged 94% (range 90-99%). Dilution linearity was demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma, and heparin plasma samples.
