Human Fucosidase Alpha L2, Plasma ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human FUCA2 in serum, plasma, tissue homogenates, or other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for FUCA2. Standards and samples are added to the wells, allowing any FUCA2 present to bind the immobilised antibody. After washing, a biotinylated anti-FUCA2 antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of an acidic stop solution. The intensity of yellow is proportional to the amount of FUCA2 captured, and concentration is calculated from the absorbance at 450 nm against a standard curve.

The kit has a stated sensitivity of 0.216 ng/ml and an assay range of 0.625-40 ng/ml, with a total assay time of 4h 30m. Detection is colorimetric and read on a microplate reader. Reported average recovery values across five samples are 91% in serum (range 80%-102%), 90% in EDTA plasma (range 81%-100%), and 84% in heparin plasma (range 80%-89%). Dilution linearity is documented at 1:2, 1:4, 1:8, and 1:16 dilutions for each of these sample types.

Kit components include a pre-coated 96-well microplate arranged as 12 x 8 well strips, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Components are stored between +4°C and -20°C as indicated for each item, and the kit should not be used past its expiration date.
