From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Free Fatty Acid Receptor 1 (also referred to as GPCR GPR40) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) with an immobilised antibody specific to the target, followed by a Biotinylated Anti-GPCR GPR40 Antibody and an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that changes to yellow upon addition of the acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader.

The quantitative assay has a working range of 1.56–100 ng/ml and a sensitivity of <0.54 ng/ml, with a total assay time of 4 hours 30 minutes. Reported spike-and-recovery values average 91% in serum (range 80–102%, n=5), 90% in EDTA plasma (81–100%, n=5) and 84% in heparin plasma (80–89%, n=5), with linearity demonstrated across 1:2 to 1:16 dilutions for each sample type. Supplied components include the pre-coated microplate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, to be stored at +4°C or -20°C as instructed.
