Human FMS Like Tyrosine Kinase 3 Ligand ELISA Kit

Manufactured for Antibodies.com as part of their Assays and Kits range, this sandwich ELISA is designed for the in vitro quantitative determination of human Flt3 Ligand/Flt3L in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) and employs the sandwich enzyme immunoassay technique, in which an antibody specific for Flt3 Ligand/Flt3L is immobilised on the plate to capture analyte from standards and test samples.

Detection is achieved using a biotinylated anti-Flt3 Ligand/Flt3L antibody and HRP-Avidin conjugate, visualised with TMB substrate; the enzymatic reaction produces a blue-coloured product that turns yellow after addition of acidic stop solution, and the concentration of Flt3 Ligand/Flt3L is calculated from the O.D. absorbance at 450 nm in a microplate reader against a standard curve. The kit shows a sensitivity of <7.5 pg/ml, an assay range of 15.6-1000 pg/ml and a total assay time of 4h 30m.

Reported average recovery is 91% in serum (n=5, range 83%-100%), 94% in EDTA plasma (n=5, range 91%-98%) and 94% in heparin plasma (n=5, range 90%-99%). Dilution linearity from 1:2 to 1:16 was demonstrated across serum (78%-104%), EDTA plasma (81%-109%) and heparin plasma (74%-105%). Kit components include the pre-coated plate, lyophilised standard, detection reagents A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored at -20°C or +4°C as instructed.
