From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human FKBP51 in serum, plasma, tissue homogenates, and other biological fluids. The assay uses a pre-coated microplate to which an antibody specific for FKBP51 has been immobilised. Standards and test samples are added to the wells, where FKBP51 is captured by the immobilised antibody. After washing, a Biotinylated Anti-FKBP51 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of FKBP51 captured, and the concentration is calculated by reading the optical density at 450nm in a microplate reader and referring to the standard curve.

The product is for research use only and is not intended for diagnostic or therapeutic use. Performance characteristics cited by the manufacturer include a sensitivity of <0.061 ng/ml, an assay range of 0.156–10 ng/ml, and a total assay time of 4h 30m. The assay is run on a colourimetric microplate reader platform with detection at 450nm.

Recovery has been evaluated in serum, EDTA plasma, and heparin plasma (n=5 for each). Reported average recoveries are 91% in serum (range 80%–102%), 90% in EDTA plasma (range 81%–100%), and 84% in heparin plasma (range 80%–89%). Linearity has been tested across dilutions of 1:2, 1:4, 1:8, and 1:16 in the same matrices, with detailed recovery ranges provided in the product documentation. Kit components should be stored at +4°C or –20°C as instructed, and the product should not be used past its expiration date.
