Human Filaggrin ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Filaggrin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips) with an immobilised anti-Filaggrin capture antibody. Standards and samples are added to the wells, followed by a biotinylated anti-Filaggrin detection antibody and an HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of the acidic stop solution; absorbance is read at 450 nm on a colorimetric microplate reader.

The product range is 0.312–20 ng/ml with a sensitivity of 0.114 ng/ml, and the total assay time is 4 hours 30 minutes. Average recovery was 91% in serum (range 80–102%, n=5), 90% in EDTA plasma (range 81–100%, n=5) and 84% in heparin plasma (range 80–89%, n=5). Sample dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma samples.

Kit components include a pre-coated 96-well microplate and lyophilised standard (2 vials) stored at −20°C, Detection Solutions A and B (120 µl each) at −20°C, 30X Wash Buffer (20 ml) at +4°C, Sample Dilution Buffer (45 ml) at −20°C, TMB Substrate (9 ml) and Stop Solution (6 ml) at +4°C, and five adhesive plate sealers. Components should be stored at +4°C or −20°C as instructed and the kit must not be used past its expiration date.
