Human Fibronectin Type III Domain Containing Protein 5 ELISA Kit

From Antibodies.com, the Human FNDC5 ELISA Kit is a sandwich enzyme-linked immunosorbent assay designed for the in vitro quantitative determination of human Fibronectin Type III Domain Containing Protein 5 (FNDC5) in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for FNDC5 immobilised on the plate. Standards and samples are incubated in the wells, followed by a biotinylated anti-FNDC5 antibody and an HRP-avidin conjugate. TMB substrate visualises the HRP enzymatic reaction; the resulting blue product turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm on a colorimetric microplate reader.

Reported sensitivity is <0.63 ng/ml with a quantitative range of 1.56–100 ng/ml. Total assay time is 4 hours 30 minutes, and kit components should be stored at +4°C or –20°C as instructed; the kit should not be used past its expiration date.

Spike-and-recovery performance (n=5) showed averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilutional linearity across 1:2, 1:4, 1:8 and 1:16 dilutions fell within the ranges: serum 74–107%, EDTA plasma 79–108%, heparin plasma 82–105%.
