From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Fibroblast Growth Factor Receptor 2 (FGFR2) in serum, plasma, tissue homogenates, and other biological fluids. The assay uses a microtiter plate pre-coated with an antibody specific for FGFR2. Standards and test samples are added to the wells and any FGFR2 present binds to the immobilised antibody. After washing, a biotinylated anti-FGFR2 detection antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of FGFR2 captured, and the concentration is calculated by reading the absorbance at 450 nm in a microplate reader against a standard curve.

The assay has a reported product range of 31.2-2000 pg/ml and a sensitivity of <12.2 pg/ml, with a total run time of 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. Recovery ranges have been validated for serum (80%-102%, average 91%, n=5), EDTA plasma (81%-100%, average 90%, n=5), and heparin plasma (80%-89%, average 84%, n=5). Linearity at 1:2, 1:4, 1:8, and 1:16 dilutions (n=5 each) is also reported; for example, at 1:2 dilution, serum measures 87-91%, EDTA plasma 90-105%, and heparin plasma 84-95%. The kit is reactive with human FGFR2.
