Human Fibroblast Growth Factor 4 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Fibroblast Growth Factor 4 (FGF4) in serum, plasma or other biological fluids. The kit employs a 96-well microtiter plate pre-coated with an antibody specific for FGF4. Standards and samples are added to the wells, where FGF4 binds to the immobilised capture antibody. Following incubation and washing, a Biotinylated Anti-FGF4 Antibody binds the captured FGF4, then HRP-Avidin conjugate is added. After further incubation and washing, TMB substrate solution produces a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow colour is proportional to the amount of FGF4 captured in each well, and concentration is calculated by reading O.D. absorbance at 450 nm against a standard curve.

The assay has a sensitivity of <14.5 pg/ml and a product range of 31.2-2000 pg/ml. It is read on a colorimetric microplate reader with a total assay time of 4h 30m. In validation testing, recovery in serum samples ranged from 80% to 102% with an average of 91% (n=5); EDTA plasma recovery ranged from 81% to 100% with an average of 90% (n=5); and heparin plasma recovery ranged from 80% to 89% with an average of 84% (n=5). Dilution linearity was demonstrated across serum, EDTA plasma and heparin plasma samples.
