Human Fibroblast Growth Factor 3 ELISA Kit

The Antibodies.com Human Fibroblast Growth Factor 3 ELISA Kit is a sandwich enzyme-linked immunosorbent assay designed for the in vitro quantitative determination of human FGF3 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microtiter plate onto which an antibody specific for FGF3 has been immobilized. Standards and samples are added to the wells, where FGF3 present in each sample is bound by the capture antibody.

After incubation and washing, a biotinylated anti-FGF3 antibody is added to bind the captured FGF3, followed by an HRP-avidin conjugate. TMB substrate solution is then used to visualize the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of an acidic stop solution. The intensity of the yellow colour is proportional to the amount of FGF3 captured in each well, and the concentration is calculated by reading the absorbance at 450nm in a colorimetric microplate reader. The reported sensitivity is 6.4 pg/ml, with a standard curve range of 15.6-1000 pg/ml. The complete assay time is approximately 4h 30m.

Validation data for the kit shows recovery ranges of 80%-102% (average 91%) for serum (n=5), 81%-100% (average 90%) for EDTA plasma (n=5), and 80%-89% (average 84%) for heparin plasma (n=5). Linear dilution testing across 1:2, 1:4, 1:8, and 1:16 dilutions produced recovery ranges of 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. Kit components are stored at either +4°C or -20°C as instructed in the protocol, and the product is intended for research use only.
