From Antibodies.com, this Human FGF2 ELISA Kit is a sandwich enzyme-linked immunosorbent assay designed for the in vitro quantitative measurement of human Fibroblast Growth Factor 2 (Basic) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated microplate with colorimetric detection read at 450 nm in a microplate reader, and the total assay time is 4 hours 30 minutes.

An antibody specific for FGF2 is immobilised on the plate to capture FGF2 from standards and test samples, followed by incubation with a Biotinylated Anti-FGF2 Antibody and then HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue product that turns yellow on addition of stop solution; the absorbance at 450 nm is proportional to the FGF2 concentration captured in each well. The standard curve spans a measuring range of 15.6-1000 pg/ml with a sensitivity of 4.55 pg/ml. In spike-and-recovery testing, serum (n=5) showed 80-102% recovery (average 91%), EDTA plasma (n=5) 81-100% (average 90%) and heparin plasma (n=5) 80-89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 gave recoveries of 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma, respectively.
