Human Fetuin B ELISA Kit

Antibodies.com provides a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human Fetuin B in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with detection performed colorimetrically by reading the O.D. absorbance at 450 nm in a microplate reader.

The reported assay range is 93.8–6000 pg/ml, with a sensitivity of less than 41.25 pg/ml, and a total assay time of 4 hours 30 minutes. The protocol uses an antibody specific for Fetuin B pre-coated onto the plate, followed by incubation with a Biotinylated Anti-Fetuin B Antibody and an HRP-Avidin conjugate; bound analyte is visualised using TMB substrate solution, producing a blue colour that turns yellow upon addition of the acidic stop solution, with yellow intensity proportional to the Fetuin B concentration.

Spike-and-recovery performance was reported in serum (n=5) at 80–102% (average 91%), EDTA plasma (n=5) at 81–100% (average 90%), and heparin plasma (n=5) at 80–89% (average 84%). Linearity was demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions in serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%). Kit components are to be stored at +4 °C or -20 °C as instructed, and the kit must not be used past its expiration date.
