From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Ferrochelatase (FECH). Reactivity is human, and the assay is intended for use with tissue homogenates, cell lysates or other biological fluids.
The assay employs a pre-coated microtiter plate. An antibody specific for FECH is immobilised on the plate; standards and samples are added, followed by a Biotinylated Anti-FECH Antibody and then HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of FECH is calculated by reference to the standard curve. The stated assay range is 0.156-10 ng/ml and sensitivity is 0.053 ng/ml. Total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
Manufacturer validation reports recovery ranges of 80%-102% for serum, 81%-100% for EDTA plasma and 80%-89% for heparin plasma, with average recovery values of 91%, 90% and 84% respectively. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 across the same three sample matrices.
