Human Fatty Acid Binding Protein 8, Myelin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com, designed for the in vitro quantitative determination of human Fatty Acid Binding Protein 8, Myelin (PMP2) in serum, plasma, tissue homogenates and other biological fluids. The assay uses the sandwich enzyme immunoassay technique, in which an antibody specific for PMP2 is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, and following incubation and washing, a Biotinylated Anti-PMP2 Antibody and an HRP-Avidin conjugate are applied sequentially. TMB substrate solution is then added, producing a blue-coloured product that changes to yellow after the addition of acidic stop solution. The intensity of yellow, read as O.D. absorbance at 450 nm, is proportional to the amount of PMP2 captured, and concentration is calculated against a standard curve.

The kit is intended for use with human samples. It has a sensitivity of <0.062 ng/ml and an assay range of 0.156–10 ng/ml. The pre-coated microplate platform consists of 12 x 8 well strips, the total assay time is 4h 30m, and the result is read on a colorimetric microplate reader. Kit components are stored at +4°C or -20°C as instructed and should not be used past the stated expiration date.

Recovery has been evaluated in serum, EDTA plasma and heparin plasma (n=5 each), with averages of 91% (range 80–102%), 90% (range 81–100%) and 84% (range 80–89%), respectively. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
