Human Fascin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Fascin in serum, plasma, tissue homogenates, cell lysates, cell culture supernatant and other biological fluids. Reactivity is human and the assay is run on a pre-coated microplate read at 450 nm using a colorimetric microplate reader.

The kit employs a sandwich enzyme immunoassay format with Biotinylated Anti-Fascin Antibody and an HRP-Avidin conjugate, visualised via TMB substrate that turns yellow after addition of acidic stop solution; the density of yellow is proportional to Fascin concentration. The stated detection range is 78.1–5000 pg/ml with a sensitivity of <34 pg/ml, and the total assay time is 4h 30m. Recovery in serum (n=5) averaged 91% (range 80–102%), in EDTA plasma 90% (81–100%) and in heparin plasma 84% (80–89%). Linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma matrices. The standard curve is generated by 1:2 serial dilution from a 5000 pg/ml stock down to 78.1 pg/ml.

Kit components include a pre-coated 96 well microplate (12 x 8 well strips), lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers, stored at +4°C or –20°C as instructed.
