Human Fas Apoptotic Inhibitory Molecule 3 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human TOSO, also known as Fas Apoptotic Inhibitory Molecule 3, in tissue homogenates, cell lysates, and other biological fluids. The kit employs a pre-coated 96-well microplate (12 x 8 well strips) with an antibody specific for TOSO. Standards and test samples are added to the wells, where TOSO is captured by the immobilized antibody. After washing, a biotinylated anti-TOSO antibody binds the captured TOSO, and an HRP-Avidin conjugate is added. TMB substrate solution produces a blue colour that changes to yellow upon addition of the acidic stop solution. The concentration of TOSO is calculated by reading the optical density at 450 nm in a microplate reader and referring to the standard curve.

The assay has a sensitivity of 0.058 ng/ml and a quantitative range of 0.156-10 ng/ml. Total assay time is approximately 4 hours 30 minutes, with colourimetric detection. Reported recovery ranges in spiked samples were 80-102% in serum (n=5, average 91%), 81-100% in EDTA plasma (n=5, average 90%), and 80-89% in heparin plasma (n=5, average 84%).
