The assay uses a 96-well microtiter plate pre-coated with an antibody specific for PANDER, with detection performed using a Biotinylated Anti-PANDER Antibody followed by an HRP-Avidin conjugate. A TMB substrate is added to visualise the enzymatic reaction, producing a coloured product that is read at 450 nm using a colorimetric microplate reader.

The kit has a sensitivity of 0.235 ng/ml and an assay range of 0.625-40 ng/ml, with a total assay time of 4 hours 30 minutes. Manufacturer-reported recovery data shows ranges of 83%-100% (average 91%) for serum (n=5), 91%-98% (average 94%) for EDTA plasma (n=5), and 90%-99% (average 94%) for heparin plasma (n=5). Linearity has been assessed across 1:2, 1:4, 1:8, and 1:16 dilutions for serum, EDTA plasma, and heparin plasma.
