An antibody specific for Adipolin is pre-coated onto a microplate. Samples and standards are added so that Adipolin is captured by the immobilised antibody. After washing, a biotinylated anti-Adipolin antibody is applied, followed by an HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of stop solution; absorbance is read at 450 nm and Adipolin concentration is determined from a standard curve. Detection is colorimetric, the working range is 0.156-10 ng/ml, the sensitivity is 0.058 ng/ml, and total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed.
