The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for ERK1 pre-coated onto a 96-well microplate. Standards and test samples are added to the wells, and captured ERK1 is detected using a Biotinylated Anti-ERK1 Antibody followed by an HRP-Avidin conjugate. Visualisation is achieved with TMB substrate solution, producing a colour change that is read at 450 nm in a microplate reader.
