Human Excitatory Amino Acid Transporter 5 ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Excitatory Amino Acid Transporter 5 (EAAT5) in tissue homogenates and other biological fluids. The assay employs a pre-coated microplate with immobilised anti-EAAT5 antibody; standards and samples are added, followed by a biotinylated anti-EAAT5 detection antibody, HRP-avidin conjugate, and TMB substrate. The enzymatic reaction produces a blue colour that changes to yellow upon addition of stop solution, and absorbance is read at 450 nm. Reported sensitivity is <0.122 ng/ml with an assay range of 0.312-20 ng/ml, and the total assay time is 4h 30m.

In addition to tissue homogenates, the kit has been evaluated with serum, EDTA plasma, and heparin plasma. Reported spike-and-recovery in serum (n=5) averaged 91% over an 80%-102% range, in EDTA plasma (n=5) averaged 90% over an 81%-100% range, and in heparin plasma (n=5) averaged 84% over an 80%-89% range. Dilution linearity has been characterised at 1:2, 1:4, 1:8, and 1:16 for each of these matrices. Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
