Human Excitatory Amino Acid Transporter 3 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Excitatory Amino Acid Transporter 3 (EAAT3) in tissue homogenates and other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for EAAT3 is immobilised on the plate; standards and test samples are added and any EAAT3 present is captured. After washing, a Biotinylated Anti-EAAT3 Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that turns yellow after addition of acidic stop solution. The concentration of EAAT3 is calculated from the O.D. absorbance read at 450 nm in a colorimetric microplate reader, by reference to the standard curve.

The kit has a sensitivity of <0.117 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4h 30m. It is intended for use with human samples and is suitable for tissue homogenates. Recovery performance has been evaluated in serum, EDTA plasma and heparin plasma (n=5 each), with average recovery ranges of 80–102% (serum, mean 91%), 81–100% (EDTA plasma, mean 90%) and 80–89% (heparin plasma, mean 84%). Dilution linearity has been tested at 1:2, 1:4, 1:8 and 1:16, with recoveries ranging from 74% to 108% across the same sample types. Kit components should be stored at +4°C or –20°C as instructed, and the product must not be used past its expiration date.
