Manufactured by Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human ERCC1 in tissue homogenates, cell lysates, and other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a pre-coated microplate. Standards and test samples are added to the wells, where ERCC1 present in each sample is bound to the wells by the immobilized antibody. Following incubation and washing, a biotinylated anti-ERCC1 antibody is added, which binds the captured ERCC1. An HRP-Avidin conjugate is then added and the plate is incubated. Following further incubation and washing, TMB substrate solution is used to visualize the HRP enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of ERCC1 captured in each well. The concentration of ERCC1 is calculated by reading the absorbance at 450nm in a microplate reader and referring to the standard curve.

The kit has a sensitivity of <0.065 ng/ml and an assay range of 0.156–10 ng/ml. The total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Recovery performance was evaluated across sample types: serum (n=5, range 80%–102%, average 91%), EDTA plasma (n=5, range 81%–100%, average 90%), and heparin plasma (n=5, range 80%–89%, average 84%). Dilutional linearity was demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma samples.
